ABSTRACT

Objective To investigate the effect of miR-17-5p on the inflammatory response and oxidative stress in the development of atherosclerosis. Methods ApoE-/ - mice were fed with high fat diet for 12 weeks to con-struct atherosclerosis (AS) model, and then miR-17-5p antagonist was injected to the caudal vein of mice subse-quently. HE staining was used to observe the pathological changes of the aorta. The apoptosis of aorta cells was de-tected by TUNEL staining. Western blot was used to detect the expression of apoptosis-related proteins and the ex-pression of inflammatory cytokines weas detected by ELISA. Oxidative stress were assessed by malondialdehyde ( MDA) levels and the activity of superoxide dismutase ( SOD) and myeloperoxidase ( MPO) . Results Inhibition of miR-17-5p alleviated the aortic lesion and apoptosis in AS mice, reduced the expression of Bax, cleaved-caspase-3 and cleaved-PARP and up-regulated the expression of Bcl-2. After inhibition of miR-17-5p, the levels of TNF-α and IL-6 in AS mice decreased, and the level of IL-10 increased. Inhibition of miR-17-5p also resulted in decrease in MDA and MPO activity, but increase in SOD activity. Conclusion Inhibition of miR-17-5p can atten-uate atherosclerotic lesions by reducing the inflammatory response and oxidative stress in the aorta of atherosclerotic mice. MiR-17-5p may be a potential target for the treatment of atherosclerosis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expression variation of RAR-β2, RASSF1A, and CDKN2A gene in the process of nickel-induced carcinogenesis.</p><p><b>METHODS</b>Nickel subsulfide (Ni(3)S(2)) at dose of 10 mg was given to Wistar rats by intramuscular injection. The mRNA expression of the three genes in induced tumors and their lung metastasis were examined by Real-time PCR. The methylation status of the 5' region of these genes were detected by Quantitative Real-time methylation specific PCR.</p><p><b>RESULTS</b>The mRNA expressions of the three genes both in muscle and lung tumor were decreased distinctly in comparison with normal tissue. But hypermethylation was found only in muscle tumor.</p><p><b>CONCLUSION</b>These findings suggest that loss of function or decrease of RAR-β2, RASSF1A, and CDKN2A, as well as the hypermethylation of 5' region of these genes, are related with nickel exposure.</p>

ABSTRACT

Objective To make a preliminary evaluation of the degradation of AZ31 bioabsorbable magnesium alloy stent implanted in the abdominal aorta of experimental rabbits. Methods Twelve AZ31 biodegradable magnesium alloy stents were separately deployed in the infrarenal abdominal aortas of twelve New Zealand white rabbits.Every three experimental rabbits were sacrificed each time at one, two, three and four months after the procedure of stenting. The stenting segment of the aortas were harvested, radiographod and sent for pathologic examination to observe the degradable performance of the stent. Results All animals survived form the operation in the scheduled follow-up period.Radiographically and pathologically, the stents were fully expanded with perfect shape one month after the procedure, and part of the stent struts began to be degraded and fractured in two months, resulting in the loss of its supporting function.Three months after the implantation most stents were corroded.and in four months all the stents become completely destroyed. The estimating time for producing complete degradation of AZ31 magnesium alloy stents in rabbit's aorta was 104.5 days. Conclusion AZ31 bioabsorbable magnesium alloy stents implanted in rabbit abdominal aorta will lose their radial force in two months. How to prolong the functioning time of the implanted stents is the next research target.

ABSTRACT

BACKGROUND: Magnalium which is potential to be the medical biodegraded metal implant is more and more interesting, but it must be well biocompatibility to human body.OBJECTIVE: To evaluate the sensitization of magnalium (AZ31B).METHODS: A total of 35 guinea pigs were randomly divided into saline group (negative control group,n=10),5% volume of formaldehyde (positive control group,n=10),and AZ31B group (n=15).Sensitization test at the maximal dosage was performed according to "Biological evaluation of medical devices-Part 10: Tests for irritation and delayed-type hypersensitivity",including intracutaneous induction, local induction, and provocation.Patch was removed after 6,24,48,and 72 hours, and the skin response was classified accordingto Magnusson and Kligman criteria.Patch was removed after 72 hours, and skin was performed with biopsy, stained with FIE staining, and observed under optic microscope.RESULTS AND CONCLUSION: Sensitization response was not tested in both negative control group and AZ31B group at 24,48,and 72 hours after patch removal; however, moderate erythema was observed in the positive control group.Optic microscope demonstrated that criteria of allergy such as spongiosis, edema, and diffuse as well as perivascular mononuclear infiltration was not observed in the AZ31B group, but a few basophilic calls ware observed. This suggested that AZ31B was biologically safe for sensitization.

ABSTRACT

Aim: To observe the influence of high fat-high sucrose diet and chronic stress on insulin resistance. Methods: Male rats were divided into four groups: control, high fat-high sucrose diet( HFSD), chronic stress( CS), high fat-high sucrose diet plus chronic stress( HFSD + CS). After feeding the animals for 10 weeks, fat, glucose and insulin concentrations in blood and PPAR-α mRNA expression in liver were examined, and glu-cose infusion rate was detected by a hyperinsulinemic euglycemic clamp experiment. Results: Insulin resistance was observed in all three treated groups, showing the highest in the HFSD + CS group. Dyslipidemia, hypergly-cosemia, hyperinsulinism and the decrease of PPAR-α mRNA expression in liver were also shown in all treated groups. There was an obvious interaction of insulin resistance, hyperglycosemia and high FFA between high fat-high sucrose diet and chronic stress. Conclusion: Combination of high fat-high sucrose diet and chronic stress could promote the development of insulin resistance, which is likely due to the high level of serum FFA.
